circRNA-disease information
Expression pattern:
UP
Associated gene: QKI, EGFR, BCL2L1, PTPN11
Associated microRNA: miR-1236-3p, miR-129-5p, miR-141-5p, miR-205-5p, miR-29a-5p, miR-942-5p
Biological function: Potential noninvasive diagnostic biomarker for breast cancer; expression was positively correlated with Ki-67 level; may participate in a circRNA-miRNA-mRNA regulatory network involved in cancer development and progression.
Molecular mechanism: Predicted circRNA-miRNA-mRNA regulatory network based on miRanda, MirTarget, STRING, Cytoscape and KEGG pathway analysis.
Biological pathway or process:

ceRNA regulation (other); autophagy (other); NF-kappaB (other); JAK/STAT (other); proliferation (other); other pathway/process (other)

Detected method:
Q
Validation methods:

RT-qPCR; Clinical Sample Validation; ROC Analysis; Bioinformatics Analysis

Clinical significance:

circ_0001640 had diagnostic potential for breast cancer, with AUC 0.7161 in the discovery cohort, 0.9802 in the validation cohort and 0.8860 in the combined cohort; its expression was positively correlated with Ki-67 level.

Description:

circ_0001640 was significantly up-regulated in whole blood from breast cancer patients and showed diagnostic value in ROC analyses. Its expression correlated positively with Ki-67 level and varied among breast cancer subtypes, with highest expression in HER2+ cancer and lowest in triple-negative cancer. The study suggests circ_0001640 may act through a predicted circRNA-miRNA-mRNA network, but mechanistic functions were not experimentally validated.

Confidence score:

0.459
