circRNA-disease information
Expression pattern:
UP
Associated gene: YAP, mut-p53, TEAD1, Pol II, aurka, mki67, bub1
Associated microRNA: miR-497-5p
Biological function: Acts as an oncogene promoting malignant phenotype and proliferation/colony formation; contributes to cisplatin resistance.
Molecular mechanism: Transcriptionally enhanced by mut-p53/YAP/TEAD complex; binds YAP and may autoregulate its own expression; directly regulates miR-497-5p via a binding site, affecting downstream proliferation genes (aurka, mki67, bub1).
Biological pathway or process:

proliferation (promotes); cell cycle (promotes); ceRNA regulation (promotes); chemoresistance (promotes); Hippo/YAP (other)

Detected method:
Q
Validation methods:

divergent primers PCR; RT-qPCR; Sanger Sequencing; RNase R Treatment; Nuclear-Cytoplasmic Fractionation; RIP (RNA Immunoprecipitation); ChIP / ChIP-seq; Transfection; Cell Cycle Assay; Colony Formation Assay; Flow Cytometry(Non-apoptosis/cycle); Bioinformatics Analysis; Survival Analysis; Western Blot

Clinical significance:

High circPVT1 levels are associated with poorer overall survival, dependent on TP53 mutations.

Description:

In HNSCC, circPVT1 is up-regulated (especially in TP53-mutant tumors) and promotes malignant phenotypes such as proliferation and colony formation, and contributes to cisplatin resistance. Mechanistically, it is transcriptionally activated by the mut-p53/YAP/TEAD complex, binds YAP (supporting an autoregulatory loop), and represses miR-497-5p to increase proliferation-associated genes (e.g., aurka, mki67, bub1). High circPVT1 expression is associated with poorer overall survival.

Confidence score:

0.8267

Other information
Title:

The oncogenic role of circPVT1 in head and neck squamous cell carcinoma is mediated through the mutant p53/YAP/TEAD transcription-competent complex.
