circRNA-disease information
Expression pattern:
DN
Associated gene: NRG1, Ago2, SREBP1, LXRa, FASN, SCD1, ACC1, CD36
Associated microRNA: miR-142-5p
Biological function: circRRM2 attenuates MASLD development, suppresses FFA-induced triglycerides accumulation, mitigates intracellular TG accumulation and lipid droplet formation, inhibits lipogenesis-related factors, and ameliorates HFD-induced steatosis and pathological changes.
Molecular mechanism: circRRM2 acts as a miR-142-5p sponge, suppresses miR-142-5p, upregulates the miR-142-5p target NRG1, and thereby inhibits lipogenesis and triglyceride accumulation in MASLD models.
Biological pathway or process:

lipid metabolism (inhibits); inflammation (inhibits); ceRNA regulation (other); other pathway/process (inhibits)

Detected method:
Q
Validation methods:

Back-Splice Junction PCR / divergent primers PCR; RNase R Treatment; Sanger Sequencing; Actinomycin D / DRB Stability Assay; RT-qPCR; Luciferase Reporter Assay; RIP (RNA Immunoprecipitation); Transfection; MTT; Oil Red O Staining; Western Blot; In Vivo Animal Model; H&E Staining; ELISA; Bioinformatics Analysis

Description:

circRRM2 is a human RRM2-derived circRNA that is downregulated in FFA-treated hepatocytes and HFD-induced MASLD mouse livers. Its overexpression alleviates MASLD-associated steatosis by sponging miR-142-5p, increasing NRG1 expression, and suppressing lipogenesis-related gene expression, triglyceride accumulation, lipid droplet formation, and inflammatory/pathological changes.

0.7712
