circRNA-disease information
Expression pattern:
UP
Associated gene: LTBP2, AGO2
Associated microRNA: miR-942-5p
Biological function: promotes OSCC cell proliferation, migration, invasion and EMT
Molecular mechanism: ceRNA sponge of miR-942-5p to increase LTBP2 expression; activates PI3K/Akt/mTOR signaling phosphorylation and accelerates EMT
Biological pathway or process:

proliferation (promotes); migration (promotes); invasion (promotes); EMT (promotes); PI3K/AKT/mTOR (promotes); ceRNA regulation (promotes)

Detected method:
Q
M
Validation methods:

Microarray; RT-qPCR; Back-Splice Junction PCR / divergent primers PCR; Sanger Sequencing; RNase R Treatment; Actinomycin D / DRB Stability Assay; FISH / smFISH; Luciferase Reporter Assay; RIP (RNA Immunoprecipitation); RNA Pull-Down; Transfection; CCK8; EdU Staining; Colony Formation Assay; Wound Healing Assay; Transwell Assay; Cell Cycle Assay; Western Blot; IF (Immunofluorescence); IHC (Immunohistochemistry); In Vivo Animal Model; ROC Analysis; Survival Analysis; Bioinformatics Analysis; Clinical Sample Validation

Clinical significance:

High expression of circEPSTI1 is associated with high T stage and advanced TNM stage and predicts poorer OS and PFS; ROC analysis indicates it discriminates OSCC from OSF and OSF from NBM

Description:

circEPSTI1 is sequentially up-regulated from NBM to OSF to OSCC and promotes OSCC cell proliferation, migration and invasion. Mechanistically, it functions as a ceRNA to sponge miR-942-5p, thereby increasing LTBP2 expression, accelerating EMT and enhancing PI3K/Akt/mTOR pathway activation; high circEPSTI1 predicts poor OS and PFS.

Confidence score:

0.8961

Other information
Title:

The circEPSTI1/mir-942-5p/LTBP2 axis regulates the progression of OSCC in the background of OSF via EMT and the PI3K/Akt/mTOR pathway.
