circRNA-disease information
Expression pattern:
UP
Associated gene: nuclear factor erythroid 2-related factor 2 (Nrf2), superoxide dismutase 2 (SOD2), histone acetyltransferase E1A binding protein p300 (EP300), Keap1
Associated microRNA: -
Biological function: CircPIBF1 promotes LUAD cell growth and tumor progression, maintains mitochondrial stability, and inhibits pyroptosis; circPIBF1 knockdown suppresses LUAD cell viability and xenograft tumor growth while enhancing pyroptosis-related factors.
Molecular mechanism: CircPIBF1 binds Nrf2 protein to promote SOD2 transcription and modulate the Nrf2-Keap1 signaling pathway; Nrf2 recruits EP300 to enhance H3K27ac modification of the SOD2 promoter, increasing SOD2 expression and suppressing ROS-mediated pyroptosis.
Biological pathway or process:

Clinical significance:

CircPIBF1 overexpression is associated with lymph node metastasis, distant metastasis, TNM stage, positive correlation with serum CEA/NSE, and shorter survival; circPIBF1 could be a prognostic biomarker and therapeutic target for LUAD.

Description:

circPIBF1 is up-regulated in lung adenocarcinoma and high expression predicts poorer survival and more aggressive clinical features. Functionally, circPIBF1 promotes LUAD cell growth and suppresses pyroptosis by binding Nrf2, enhancing SOD2 transcription, and facilitating EP300-mediated H3K27ac modification of the SOD2 promoter. Knockdown of circPIBF1 inhibits tumor growth in vitro and in xenograft models, supporting its potential as a prognostic biomarker and therapeutic target.
