circ-GLI1 (hsa_circ_0027247), derived from GLI1, is up-regulated in melanoma cells and promotes migration/invasion, metastasis, and angiogenesis. Mechanistically, it binds p70S6K2 to increase GSK3beta Ser9 phosphorylation, stabilizing GLI1 and beta-catenin proteins to activate Hedgehog/GLI1 and Wnt/beta-catenin signaling, thereby inducing MYC-driven CYR61 transcription.
