circRNA_100284 (from host gene GCLM) is upregulated during arsenite-induced malignant transformation of human hepatic epithelial cells and is enriched in exosomes released by transformed cells. Exosomal circRNA_100284 is transferred to neighboring non-transformed cells, where it sponges miR-217, leading to increased EZH2/cyclin-D1, accelerated G1/S transition, enhanced proliferation, and promotion of malignant phenotypes. Circulating exosomal circRNA_100284 is increased in sera of arsenite-exposed individuals, suggesting biomarker potential for arsenite exposure/arsenicosis. |
