circRNA-disease information
Expression pattern:
UP
Associated gene: HMGA2, Twist1, Ago2
Associated microRNA: miR-370
Biological function: promotes proliferation, migration, invasion, and tumor growth in lung adenocarcinoma
Molecular mechanism: circASPH is transcriptionally upregulated by HMGA2/Twist1 and functions as a ceRNA sponge for miR-370 to relieve repression of HMGA2, forming an HMGA2-circASPH-HMGA2 positive feedback axis
Biological pathway or process:

proliferation (promotes); migration (promotes); invasion (promotes); EMT (promotes); ceRNA regulation (promotes)

Detected method:
Q
S
Validation methods:

circRNA-seq; Bioinformatics Analysis; RT-qPCR; RNase R Treatment; FISH / smFISH; RIP (RNA Immunoprecipitation); Luciferase Reporter Assay; RNA Pull-Down; Transfection; EdU Staining; Wound Healing Assay; Transwell Assay; In Vivo Animal Model; Western Blot; IHC (Immunohistochemistry); ChIP / ChIP-seq; Co-IP; Survival Analysis; Clinical Sample Validation

Clinical significance:

circASPH level was positively correlated with T and N stages; higher circASPH predicted shorter overall survival

Description:

circASPH is up-regulated in lung adenocarcinoma tissues and cell lines and is associated with more advanced T/N stages and shorter overall survival. It promotes proliferation, migration, invasion, and xenograft tumor growth. Mechanistically, it is transcriptionally regulated by HMGA2 and Twist1 and sponges miR-370 to relieve repression of HMGA2, forming an HMGA2-circASPH-HMGA2 positive feedback axis.

Confidence score:

0.8775
