circRNA-disease information
Expression pattern:
UP
Associated gene: HuR, NUCKS1, hnRNPA2B1
Associated microRNA: -
Biological function: promotes proliferation, invasion, migration, tumor growth and metastasis; promotes extracellular vesicles-mediated macrophage M2 polarization
Molecular mechanism: forms an RNA-protein complex with HuR to increase NUCKS1 expression; activates PI3K/AKT/mTOR signaling; hnRNPA2B1 mediates packaging of circATP9A into extracellular vesicles that induce TAM M2 polarization
Biological pathway or process:

PI3K/AKT/mTOR (promotes); proliferation (promotes); migration (promotes); invasion (promotes); metastasis (promotes); macrophage polarization (promotes); immune regulation (other); ceRNA regulation (other)

Validation methods:

Microarray; RT-qPCR; RNase R Treatment; Sanger Sequencing; divergent primers PCR; Gel Electrophoresis; ISH (In Situ Hybridization); FISH / smFISH; RNA Pull-Down; RIP (RNA Immunoprecipitation); Western Blot; Transfection; CCK8; EdU Staining; Colony Formation Assay; Transwell Assay; In Vivo Animal Model; IHC (Immunohistochemistry); IF (Immunofluorescence); Flow Cytometry(Non-apoptosis/cycle); Bioinformatics Analysis

Clinical significance:

circATP9A is upregulated in NSCLC tissues and higher in advanced tumor stages; proposed as a diagnostic indicator and therapeutic target

Description:

circATP9A is up-regulated in NSCLC and promotes proliferation, migration/invasion, tumor growth and metastasis. Mechanistically, it binds the RBP HuR to increase NUCKS1 mRNA/protein and activate PI3K/AKT/mTOR signaling, and it is packaged into extracellular vesicles via hnRNPA2B1 to drive TAM M2 polarization, promoting NSCLC progression.

Confidence score:

0.8446

Other information
Title:
