circRNA-disease information
Expression pattern:
UP
Associated gene: FOSL2, PCBP1, CD44v6, AGO2
Associated microRNA: miR-143-3p
Biological function: promotes EMT, migration, invasion, proliferation and metastasis in HCC
Molecular mechanism: Acts as a ceRNA sponging miR-143-3p to derepress FOSL2; binds the RBP PCBP1 to increase CD44v6 expression
Biological pathway or process:

EMT (promotes); migration (promotes); invasion (promotes); proliferation (promotes); metastasis (promotes); ceRNA regulation (promotes)

Detected method:
Q
H
S
Validation methods:

RNA-seq; RT-qPCR; ISH (In Situ Hybridization); Back-Splice Junction PCR / divergent primers PCR; Sanger Sequencing; RNase R Treatment; FISH / smFISH; Nuclear-Cytoplasmic Fractionation; RNA Pull-Down; RIP (RNA Immunoprecipitation); Luciferase Reporter Assay; Transfection; CCK8; Transwell Assay; Wound Healing Assay; Western Blot; Clinical Sample Validation; Cohort Study; In Vivo Animal Model; H&E Staining

Clinical significance:

High circ0003998 expression correlates with advanced TNM stage and high serum AFP; reported as an independent prognostic factor for poor OS in HCC (from authors' previous study).

Description:

circ0003998 (hsa_circ_0003998), derived from ARFGEF2, is up-regulated in HCC and PVTT tissues and promotes EMT, migration/invasion, proliferation and metastasis. Mechanistically it sponges miR-143-3p to derepress FOSL2 and also binds the RBP PCBP1 to increase CD44v6 expression, forming circ0003998/miR-143-3p/FOSL2 and circ0003998/PCBP1/CD44v6 axes.

Confidence score:

0.8689
