circRNA-disease information
Expression pattern:
UP
Associated gene: ZEB2
Associated microRNA: miR-377
Biological function: Promotes bladder cancer cell growth/proliferation, migration and invasion; arrests cell cycle at G0/G1 when silenced; promotes apoptosis when silenced.
Molecular mechanism: Acts as a miRNA sponge for miR-377, thereby promoting ZEB2 expression; ZEB2 restoration rescues the suppressive effects of circZFR knockdown.
Biological pathway or process:

proliferation (promotes); migration (promotes); invasion (promotes); apoptosis (inhibits); cell cycle (promotes); ceRNA regulation (promotes)

Detected method:
Q
Validation methods:

RT-qPCR; Clinical Sample Validation; Cohort Study; Survival Analysis; ROC Analysis; Bioinformatics Analysis; RNA Pull-Down; Luciferase Reporter Assay; Transfection; MTT; Colony Formation Assay; Cell Cycle Assay; Annexin V/PI Flow Cytometry; Transwell Assay; Western Blot; IHC (Immunohistochemistry)

Clinical significance:

Higher circZFR levels were positively correlated with pathological T stage, grade, lymphatic metastasis, recurrence, and poorer progression-free survival (PFS) and overall survival (OS); ROC suggested diagnostic value (AUC = 0.8216).

Description:

CircZFR is up-regulated in bladder cancer tissues and cell lines and is associated with worse PFS/OS. Functionally, circZFR promotes proliferation, migration and invasion and inhibits apoptosis. Mechanistically, it acts as a ceRNA by sponging miR-377 to upregulate ZEB2, and ZEB2 restoration rescues the effects of circZFR knockdown.

Confidence score:

0.7929
