circRNA-disease information
Expression pattern:
UP
Associated gene: Beclin1, RUNX1, ATG13
Associated microRNA: miR-199a-5p / miR-199a
Biological function: promotes autophagy flux, proliferation, invasion and metastasis in EOC
Molecular mechanism: ceRNA sponging miR-199a-5p to de-repress Beclin1 and RUNX1; RUNX1 transcriptionally elevates circMUC16; circMUC16 directly binds ATG13 to promote its expression
Biological pathway or process:

autophagy (promotes); proliferation (promotes); invasion (promotes); metastasis (promotes); ceRNA regulation (promotes)

Detected method:
Q
S
Validation methods:

RNA-seq; RT-qPCR; FISH / smFISH; RNA Pull-Down; Luciferase Reporter Assay; Transfection; EdU Staining; Transwell Assay; Western Blot; IHC (Immunohistochemistry); ChIP / ChIP-seq; In Vivo Animal Model; Bioinformatics Analysis; Clinical Sample Validation

Clinical significance:

circMUC16 expression was linked to EOC stage and grade; potential target for EOC diagnosis and therapy

Description:

circMUC16 (hsa_circ_0049116), derived from MUC16, is up-regulated in EOC tissues and patient serum. It promotes EOC autophagy flux and malignant phenotypes (proliferation, invasion/metastasis) by sponging miR-199a-5p to de-repress Beclin1 and RUNX1, forms a RUNX1-circMUC16 positive feedback loop, and also directly binds ATG13 to enhance its expression.

Confidence score:

0.8023
