circRNA-disease information
Expression pattern:
UP
Associated gene: SGPP1, CyclinD1, Caspase-3
Associated microRNA: miR-525-5p
Biological function: circ_0000423 promotes CRC cell proliferation, migration, invasion and tumorigenesis, inhibits apoptosis, and counteracts the inhibitory effects of sevoflurane on CRC progression.
Molecular mechanism: circ_0000423 functions as a ceRNA by sponging miR-525-5p to positively regulate SGPP1 expression, thereby mediating sevoflurane-regulated CRC progression.
Biological pathway or process:

proliferation (promotes); migration (promotes); invasion (promotes); apoptosis (inhibits); ceRNA regulation (other); other pathway/process (promotes)

Detected method:
Q
Validation methods:

Back-Splice Junction PCR / divergent primers PCR; RT-qPCR; Clinical Sample Validation; Bioinformatics Analysis; Luciferase Reporter Assay; RIP (RNA Immunoprecipitation); Transfection; CCK8; Colony Formation Assay; Annexin V/PI Flow Cytometry; Transwell Assay; Western Blot; In Vivo Animal Model; IHC (Immunohistochemistry)

Clinical significance:

circ_0000423 was upregulated in CRC tumor tissues and was proposed as an effective therapeutic target for CRC.

Description:

circ_0000423 is upregulated in colorectal cancer tissues and cells and is reduced by sevoflurane treatment. Functionally, circ_0000423 promotes CRC proliferation, migration, invasion and tumor growth while suppressing apoptosis, mainly through the circ_0000423/miR-525-5p/SGPP1 ceRNA axis. Overexpression of circ_0000423 weakens the anti-tumor effects of sevoflurane, suggesting its potential relevance as a therapeutic target in CRC.

Confidence score:

0.8005
