circRNA-disease information
Expression pattern:
UP
Associated gene: HuR, KLF2, RNF128
Associated microRNA: -
Biological function: Promotes PC cell proliferation, invasion, migration, tumorigenesis, and lung/liver metastasis.
Molecular mechanism: Tumor-derived EVs shuttle circFMN2 into PC cells; circFMN2 binds HuR, decreases HuR-KLF2 interaction, destabilizes KLF2 mRNA, represses KLF2 expression, reduces KLF2 binding to the RNF128 promoter, and represses RNF128 transcription.
Biological pathway or process:

proliferation (promotes); invasion (promotes); migration (promotes); metastasis (promotes); mRNA stability (inhibits); other pathway/process (other)

Detected method:
Q
Validation methods:

RNase R Treatment; Actinomycin D / DRB Stability Assay; RT-qPCR; FISH / smFISH; IF (Immunofluorescence); Clinical Sample Validation; RIP (RNA Immunoprecipitation); RNA Pull-Down; Co-IP; ChIP / ChIP-seq; Luciferase Reporter Assay; Transfection; CCK8; Colony Formation Assay; Transwell Assay; In Vivo Animal Model; H&E Staining; IHC (Immunohistochemistry); Western Blot; Bioinformatics Analysis

Clinical significance:

circFMN2 expression was associated with tumor stage, lymph node metastasis, and Gleason score; PC-EVs-derived circFMN2 may serve as a potential diagnostic and therapeutic molecular target.

Description:

circFMN2 is upregulated in prostatic cancer tissues and cell lines and is shuttled between PC cells by tumor-derived extracellular vesicles. It promotes proliferation, invasion, migration, tumor growth, and lung/liver metastasis by binding HuR, reducing HuR-mediated KLF2 mRNA stability, and repressing the KLF2/RNF128 transcriptional axis. Clinically, higher circFMN2 expression correlates with tumor stage, lymph-node metastasis, and Gleason score.

Confidence score:

0.8644
