circRNA-disease information
Expression pattern:
UP
Associated gene: E2F1, ESRP1, Cyclin E, Pre-CAMSAP1, Ago2
Associated microRNA: miR-328-5p
Biological function: Promotes malignant behavior, CRC cell proliferation, G1/S cell cycle progression and tumor growth; functions as an oncogenic circRNA and biomarker candidate.
Molecular mechanism: ESRP1-mediated circCAMSAP1 biogenesis; circCAMSAP1 acts as a cytoplasmic miR-328-5p sponge, relieving miR-328-5p-mediated suppression of E2F1 and increasing Cyclin E expression.
Biological pathway or process:

proliferation (promotes); cell cycle (promotes); apoptosis (other); ceRNA regulation (promotes); mRNA stability (other); other pathway/process (promotes)

Detected method:

S
Validation methods:

Back-Splice Junction PCR / divergent primers PCR; RNase R Treatment; Sanger Sequencing; Actinomycin D / DRB Stability Assay; RT-qPCR; RNA-seq; FISH / smFISH; ISH (In Situ Hybridization); IHC (Immunohistochemistry); IF (Immunofluorescence); Nuclear-Cytoplasmic Fractionation; Clinical Sample Validation; RIP (RNA Immunoprecipitation); RNA Pull-Down; Luciferase Reporter Assay; Transfection; EdU Staining; Colony Formation Assay; Annexin V/PI Flow Cytometry; Cell Cycle Assay; Flow Cytometry(Non-apoptosis/cycle); In Vivo Animal Model; H&E Staining; Western Blot; Cohort Study; Survival Analysis; ROC Analysis; Bioinformatics Analysis; Droplet digital PCR

Clinical significance:

Increased circCAMSAP1 expression was correlated with advanced TNM stage and shortened overall survival; circCAMSAP1 may serve as a diagnostic and prognostic biomarker for CRC, including detection in serum before surgery.

Description:

circCAMSAP1 is an up-regulated CAMSAP1-derived circRNA in colorectal cancer. It promotes CRC cell proliferation and tumor growth by sponging miR-328-5p and relieving repression of E2F1, while its biogenesis is promoted by ESRP1. Clinically, high circCAMSAP1 expression is associated with advanced stage and poor overall survival, and serum circCAMSAP1 may have diagnostic/prognostic biomarker potential.

Confidence score:

0.901
