| Associated gene: |
RUNX1, Y-box binding protein-1 (YBX1), YBX1, EGFP, c-Met, CDC25A |
| Associated microRNA: |
miR-548g |
| Biological function: |
Acts as a tumor suppressor in gastric cancer; overexpression inhibits GC cell proliferation, migration, invasion, colony formation and xenograft tumor growth. |
| Molecular mechanism: |
circFAT1(e2) acts as a miR-548g sponge in the cytoplasm to release/upregulate RUNX1, and nuclear circFAT1(e2) directly interacts with YBX1 to inhibit YBX1 function and downstream targets. |
| Biological pathway or process: |
proliferation (inhibits); migration (inhibits); invasion (inhibits); metastasis (inhibits); ceRNA regulation (promotes); other pathway/process (inhibits) |

| Validation methods: |
Back-Splice Junction PCR / divergent primers PCR; RNase R Treatment; Sanger Sequencing; Actinomycin D / DRB Stability Assay; RT-qPCR; Microarray; FISH / smFISH; Clinical Sample Validation; RIP (RNA Immunoprecipitation); RNA Pull-Down; Luciferase Reporter Assay; Transfection; MTT; Colony Formation Assay; Transwell Assay; In Vivo Animal Model; IHC (Immunohistochemistry); Western Blot; Survival Analysis; Bioinformatics Analysis |
| Clinical significance: |
High circFAT1(e2) expression predicts better prognosis and may serve as a prognostic biomarker for GC patients; expression is correlated with pN status, pM status and clinical stage. |
| Description: |
circFAT1(e2), also identified as hsa_circ_0001461, is a FAT1-derived circular RNA that is downregulated in gastric cancer tissues and cell lines. It functions as a tumor suppressor by sponging miR-548g to upregulate RUNX1 in the cytoplasm and by directly interacting with YBX1 in the nucleus, thereby suppressing proliferation, migration, invasion and xenograft tumor growth. Higher circFAT1(e2) expression is associated with better prognosis in GC patients. |
