| Expression pattern: |
DN |
| Associated gene: |
ITCH, Notch1, Wnt/beta-catenin signaling pathways, Dvl3, c-myc |
| Associated microRNA: |
miR-7, miR-24, miR-124 |
| Biological function: |
inhibits cell proliferation; promotes apoptosis |
| Molecular mechanism: |
May act as a miRNA sponge (miR-7/miR-24/miR-124) and may regulate Notch1 and Wnt/beta-catenin signaling; may downregulate phosphorylated Dvl3 and inhibit c-myc expression. |
| Biological pathway or process: |
proliferation (inhibits); apoptosis (promotes); Wnt/beta-catenin (inhibits) |

| Validation methods: |
RT-qPCR; Clinical Sample Validation; ROC Analysis; Survival Analysis; Transfection; CCK8; Annexin V/PI Flow Cytometry |
| Clinical significance: |
circ-ITCH could distinguish MM patients from healthy controls (AUC 0.809); high circ-ITCH expression correlated with decreased ISS stage and prolonged PFS and OS. |
| Description: |
circ-ITCH is down-regulated in multiple myeloma bone marrow samples and in several MM cell lines. Higher circ-ITCH expression is associated with lower ISS stage and longer PFS/OS, and circ-ITCH overexpression suppresses proliferation while promoting apoptosis in U226 cells, suggesting a tumor-suppressive role and potential biomarker/therapeutic value in MM. |
