circMVP (hsa_circ_0000688) is up-regulated in colorectal cancer and promotes tumor growth, invasion and immune evasion. Mechanistically, circMVP binds and stabilizes METTL3 (mainly in the nucleus), enhances METTL3-mediated m6A modification on CTNNB1 mRNA to increase beta-catenin translation/nuclear activity, and induces CD276/B7-H3 expression to suppress anti-tumor T cell responses. Targeting circMVP enhances the efficacy of anti-B7-H3 immunotherapy in vivo. |
