circRNA-disease information
Expression pattern:
UP
Associated gene: KLF6
Associated microRNA: miR-122-5p
Biological function: circ_FOXO3 suppresses H2O2-induced HBVSMC proliferation, promotes apoptosis, aggravates H2O2-induced HBVSMC damage, and is implicated in IA development.
Molecular mechanism: circ_FOXO3 acts as a sponge for miR-122-5p and regulates the miR-122-5p/KLF6 axis to modulate H2O2-induced HBVSMC proliferation and apoptosis.
Biological pathway or process:

ceRNA regulation (promotes); proliferation (inhibits); apoptosis (promotes); other pathway/process (promotes)

Detected method:
Q
Validation methods:

RT-qPCR; Clinical Sample Validation; Bioinformatics Analysis; Luciferase Reporter Assay; RNA Pull-Down; RIP (RNA Immunoprecipitation); Transfection; MTT; Colony Formation Assay; Annexin V/PI Flow Cytometry; Western Blot

Clinical significance:

circ_FOXO3 expression was associated with smoking history, Hunt-Hess grading, and endothelial injury degree in IA patients; the circ_FOXO3/miR-122-5p/KLF6 axis was proposed as a promising treatment approach.

Description:

This study found that circ_FOXO3 is up-regulated in IA tissues and in H2O2-treated HBVSMC. Functionally, circ_FOXO3 inhibits HBVSMC proliferation and promotes apoptosis, thereby aggravating H2O2-induced HBVSMC damage relevant to IA. Mechanistically, circ_FOXO3 sponges miR-122-5p and regulates its target KLF6, forming a circ_FOXO3/miR-122-5p/KLF6 axis with potential therapeutic significance.

Confidence score:

0.7277

Other information
Title:

Circ_FOXO3 regulates KLF6 through sponge adsorption of miR-122-5p to repress H2O2-induced HBVSMC proliferation, thus promoting IA development in vitro model.
