circRNA-disease information
Expression pattern:
UP
Associated gene: GCLM, Nrf2, GPX4, xCT, GCLC, FTH1
Associated microRNA: miR-145-5p
Biological function: Promotes MMQ cell proliferation and tumor growth, attenuates cabergoline efficacy, and induces resistance to CAB-induced ferroptosis in prolactinoma models.
Molecular mechanism: circOMA1 acts as a miR-145-5p sponge to regulate GCLM expression and inhibits ferroptosis through promotion of the Nrf2/GPX4 axis, thereby reducing cabergoline efficacy.
Biological pathway or process:

proliferation (promotes); ferroptosis (inhibits); drug resistance (promotes); ceRNA regulation (other); other pathway/process (promotes)

Detected method:
Q
Validation methods:

RT-qPCR; Clinical Sample Validation; Transfection; CCK8; Colony Formation Assay; EdU Staining; Western Blot; IF (Immunofluorescence); IHC (Immunohistochemistry); Luciferase Reporter Assay; Bioinformatics Analysis; In Vivo Animal Model

Clinical significance:

High circOMA1 expression is associated with DA-resistant prolactinoma and elevated prolactin levels, and circOMA1 may be a therapeutic target for individualized treatment of DA-resistant prolactinoma patients.

Description:

circOMA1 is up-regulated in DA-resistant prolactinoma tissues and promotes prolactinoma cell growth in vitro and in a xenograft model. Mechanistically, circOMA1 acts as a miR-145-5p sponge to regulate GCLM and promotes the Nrf2/GPX4 ferroptosis-resistance program, thereby attenuating cabergoline-induced ferroptosis and reducing cabergoline efficacy.

Confidence score:

0.713
