circRNA-disease information
Expression pattern:
UP
Associated gene: HLF
Associated microRNA: miR-200b-3p
Biological function: circCHD2 promotes TGF-beta1-induced LX-2 cell proliferation and increases liver fibrosis markers alpha-SMA and Col1A1; circCHD2 knockdown suppresses activated HSC proliferation and reduces alpha-SMA and Col1A1 levels.
Molecular mechanism: circCHD2 functions as a ceRNA by sponging miR-200b-3p to regulate downstream HLF in liver cirrhosis-related activated HSCs.
Biological pathway or process:

proliferation (promotes); fibrosis (promotes); ceRNA regulation (promotes)

Detected method:
Q
Validation methods:

RT-qPCR; Clinical Sample Validation; Bioinformatics Analysis; Luciferase Reporter Assay; Transfection; CCK8; EdU Staining; Colony Formation Assay; Western Blot

Clinical significance:

circCHD2 may have potential to be a therapeutic target of liver cirrhosis.

Description:

In this study, circCHD2 was up-regulated in serum from patients with liver cirrhosis and in TGF-beta1-activated LX-2 hepatic stellate cells. Functionally, circCHD2 promoted activated HSC proliferation and fibrosis-marker expression, while circCHD2 knockdown alleviated these effects through the miR-200b-3p/HLF ceRNA axis. The authors propose circCHD2 as a potential therapeutic target for liver cirrhosis.
