circRNA-disease information
Expression pattern:
UP
Associated gene: IGF2BP1, CXCL5 mRNA
Associated microRNA: -
Biological function: Promotes Ang II-induced HA-VSMCs proliferation, migration and phenotypic switch; contributes to TAD progression.
Molecular mechanism: circNRIP1 recruits/binds IGF2BP1 and increases CXCL5 mRNA stability, thereby promoting CXCL5 expression.
Biological pathway or process:

proliferation (promotes); migration (promotes); mRNA stability (promotes); other pathway/process (promotes)

Detected method:
Q
M
Validation methods:

Back-Splice Junction PCR / divergent primers PCR; RNase R Treatment; RT-qPCR; Nuclear-Cytoplasmic Fractionation; Clinical Sample Validation; RIP (RNA Immunoprecipitation); Actinomycin D / DRB Stability Assay; Transfection; EdU Staining; Transwell Assay; Wound Healing Assay; Western Blot; Bioinformatics Analysis

Clinical significance:

High circNRIP1 expression was associated with hypertension and D-dimer of TAD patients; targeting circNRIP1/IGF2BP1/CXCL5 axis may be an effective measure for TAD treatment.

Description:

circNRIP1 is up-regulated in TAD patient aortic specimens and Ang II-induced HA-VSMCs. It promotes HA-VSMC proliferation, migration and phenotypic switch by binding/recruiting the RBP IGF2BP1 to enhance CXCL5 mRNA stability, suggesting the circNRIP1/IGF2BP1/CXCL5 axis as a potential therapeutic target for TAD.

Confidence score:

0.7968

Other information
