circRNA-disease information
Expression pattern:
UP
Associated gene: MYC, BCL2, YAP1, POU2F3
Associated microRNA: -
Biological function: circPVT1 promotes lung cell proliferation, reduces apoptosis, increases MYC and BCL2 expression, modulates SCLC subtype-related master regulators by increasing YAP1 and decreasing POU2F3, and is secreted in extracellular vesicles from SCLC cell lines.
Molecular mechanism: circPVT1 overexpression induces transcriptional and protein-level changes involving MYC, BCL2, YAP1, POU2F3, proliferation factors, and apoptosis-related genes; it is unlikely to encode a protein product and can be packaged into extracellular vesicles.
Biological pathway or process:

proliferation (promotes); cell cycle (promotes); apoptosis (inhibits); Hippo/YAP (promotes); other pathway/process (other)

Detected method:

Validation methods:

RNase R Treatment; RT-qPCR; Northern Blot; Transfection; Flow Cytometry(Non-apoptosis/cycle); Annexin V/PI Flow Cytometry; Western Blot; Bioinformatics Analysis

Clinical significance:

Potential biomarker for MYC-driven SCLC and noninvasive liquid biopsy-based approaches, based on detection in extracellular vesicles secreted by SCLC cell lines.

Description:

This study reports that circPVT1 is overexpressed in SCLC cell lines, especially MYC-amplified contexts. Functional overexpression experiments indicate that circPVT1 promotes proliferation, inhibits apoptosis, increases MYC/BCL2/YAP1, decreases POU2F3, and may contribute to SCLC tumorigenesis and subtype definition. circPVT1 is also detected in extracellular vesicles from SCLC cell lines, supporting potential biomarker relevance.

Confidence score:

0.593
