circPTTG1IP is a human chromosome 21-derived tumor suppressor circRNA that is downregulated in HCC and whose low expression predicts poor prognosis. It suppresses HCC malignancy by sponging miR-16-5p to increase RNF125, leading to ubiquitination and degradation of JAK1 and inhibition of JAK1-STAT3 signaling. Loss of circPTTG1IP promotes tumor progression and macrophage recruitment/M2 polarization, while the JAK1 inhibitor filgotinib counteracts these effects. |
