circRNA-disease information
Expression pattern:
UP
Associated gene: CXCR4
Associated microRNA: miR-373-3p
Biological function: circ_0001721 aggravates IL-1beta-induced chondrocyte injury by promoting chondrocyte apoptosis, extracellular matrix degradation and inflammation, while suppressing proliferation; circ_0001721 knockdown alleviates these injury phenotypes.
Molecular mechanism: circ_0001721 functions as a miR-373-3p sponge to upregulate CXCR4, thereby aggravating IL-1beta-induced chondrocyte injury.
Biological pathway or process:

ceRNA regulation (other); proliferation (inhibits); apoptosis (promotes); inflammation (promotes); other pathway/process (promotes)

Detected method:
Q
Validation methods:

Back-Splice Junction PCR / divergent primers PCR; RNase R Treatment; Actinomycin D / DRB Stability Assay; RT-qPCR; Nuclear-Cytoplasmic Fractionation; Clinical Sample Validation; RIP (RNA Immunoprecipitation); RNA Pull-Down; Luciferase Reporter Assay; Transfection; MTT; EdU Staining; Annexin V/PI Flow Cytometry; ELISA; Western Blot; Bioinformatics Analysis

Clinical significance:

circ_0001721 is highly expressed in OA patients and might be a potential therapeutic target for OA.

Description:

In this study, circ_0001721 was up-regulated in cartilage tissues from OA patients and in IL-1beta-induced human chondrocytes. Functional experiments showed that circ_0001721 knockdown relieved IL-1beta-induced chondrocyte injury by promoting proliferation and reducing apoptosis, ECM degradation and inflammation. Mechanistically, circ_0001721 acted as a sponge for miR-373-3p to upregulate CXCR4, supporting its potential as a therapeutic target for OA.

Confidence score:

0.8417
