circRNA-disease information
Expression pattern:
UP
Associated gene: ANLN, CyclinD1, Cleaved-caspase3, MMP9, Ki67
Associated microRNA: miR-200b-3p
Biological function: Contributes to PTX resistance, promotes proliferation, cell cycle progression, migration and invasion, suppresses apoptosis, and promotes xenograft tumor growth in breast cancer.
Molecular mechanism: circ_0,007,331 functions as a miRNA sponge for miR-200b-3p and positively regulates ANLN expression, thereby mediating PTX resistance and malignant phenotypes of breast cancer cells.
Biological pathway or process:

chemoresistance (promotes); drug resistance (promotes); proliferation (promotes); migration (promotes); invasion (promotes); apoptosis (inhibits); cell cycle (promotes); ceRNA regulation (promotes)

Detected method:
Q
Validation methods:

RNase R Treatment; Actinomycin D / DRB Stability Assay; RT-qPCR; Clinical Sample Validation; Transfection; CCK8; Cell Cycle Assay; Annexin V/PI Flow Cytometry; Transwell Assay; Western Blot; Bioinformatics Analysis; Luciferase Reporter Assay; RNA Pull-Down; In Vivo Animal Model; IHC (Immunohistochemistry)

Clinical significance:

circ_0,007,331 might be a potential PTX-resistant biomarker in BC cells; combined therapy of circ_0,007,331 knockdown and PTX treatment may be effective for nontriple negative BC patients.

Description:

circ_0,007,331 is up-regulated in breast cancer tissues and PTX-resistant breast cancer cell lines. It promotes PTX resistance, proliferation, cell cycle progression, migration, invasion and xenograft tumor growth while inhibiting apoptosis, at least partly by sponging miR-200b-3p to increase ANLN expression.

Confidence score:

0.8298
