circRNA-disease information
Expression pattern:
UP
Associated gene: NFIB
Associated microRNA: miR-146b-5p
Biological function: circ_VCAN promotes glioma cell proliferation, migration and invasion, suppresses apoptosis, and attenuates the inhibitory effect of sevoflurane on glioma tumor growth.
Molecular mechanism: circ_VCAN acts as a competing endogenous RNA sponge for miR-146b-5p, thereby upregulating NFIB expression and mediating sevoflurane-related inhibition of glioma malignant phenotypes.
Biological pathway or process:

proliferation (promotes); migration (promotes); invasion (promotes); apoptosis (inhibits); ceRNA regulation (promotes); other pathway/process (other)

Detected method:
Q
Validation methods:

RT-qPCR; Actinomycin D / DRB Stability Assay; Nuclear-Cytoplasmic Fractionation; Clinical Sample Validation; Luciferase Reporter Assay; Transfection; CCK8; MTT; EdU Staining; Colony Formation Assay; Annexin V/PI Flow Cytometry; Transwell Assay; Western Blot; IHC (Immunohistochemistry); In Vivo Animal Model; Bioinformatics Analysis

Clinical significance:

circ_VCAN was apparently upregulated in glioma tissues relative to normal tissues, suggesting potential disease relevance, but no diagnostic or prognostic performance was reported.

Description:

circ_VCAN is upregulated in glioma tissues and is downregulated by sevoflurane treatment in glioma cells. Functionally, circ_VCAN overexpression counteracts sevoflurane-mediated inhibition of proliferation, migration, invasion and tumor growth, while reducing sevoflurane-induced apoptosis. Mechanistically, circ_VCAN sponges miR-146b-5p to upregulate NFIB, forming a circ_VCAN/miR-146b-5p/NFIB axis in glioma.

Confidence score:

0.7861
