circRNA-disease information
Expression pattern:
UP
Associated gene: TNFSF11
Associated microRNA: miR-149-5p
Biological function: circ_CDYL promotes Ang II-induced podocyte apoptosis; knockdown of circ_CDYL inhibits apoptosis in podocytes and protects against Ang II-induced injury in the MN model.
Molecular mechanism: circ_CDYL functions through a ceRNA-like mechanism by interacting with miR-149-5p and modulating TNFSF11 expression, thereby regulating Ang II-induced podocyte apoptosis.
Biological pathway or process:

apoptosis (promotes); ceRNA regulation (promotes)

Detected method:
Q
Validation methods:

RT-qPCR; RNase R Treatment; Actinomycin D / DRB Stability Assay; Clinical Sample Validation; Transfection; Annexin V/PI Flow Cytometry; Luciferase Reporter Assay; Western Blot; ROC Analysis; Bioinformatics Analysis

Clinical significance:

Serum circ_CDYL may serve as a new non-invasive and specific marker for MN patients, with an AUC of 0.8600.

Description:

circ_CDYL is up-regulated in renal tissues and serum from MN patients and in Ang II-induced human podocytes. Functionally, circ_CDYL promotes podocyte apoptosis, while circ_CDYL knockdown protects against Ang II-induced apoptosis. Mechanistically, circ_CDYL interacts with miR-149-5p to regulate TNFSF11 expression, and serum circ_CDYL shows potential diagnostic value for MN.

Confidence score:

0.7905
