circRNA-disease information
Expression pattern:
UP
Associated gene: DDIT4
Associated microRNA: miR-495-3p
Biological function: promotes MM cell proliferation, reduces apoptosis, and accelerates cell cycle progression; promotes tumor growth in vivo
Molecular mechanism: ceRNA mechanism: circXPO1 sponges miR-495-3p to increase DDIT4 expression
Biological pathway or process:

proliferation (promotes); apoptosis (inhibits); cell cycle (promotes); ceRNA regulation (promotes)

Detected method:
Q
M
Validation methods:

Microarray; RT-qPCR; RNase R Treatment; Actinomycin D / DRB Stability Assay; Nuclear-Cytoplasmic Fractionation; Clinical Sample Validation; ROC Analysis; Survival Analysis; RNA Pull-Down; Luciferase Reporter Assay; Bioinformatics Analysis; Transfection; CCK8; EdU Staining; Annexin V/PI Flow Cytometry; Cell Cycle Assay; Western Blot; In Vivo Animal Model; H&E Staining; IHC (Immunohistochemistry)

Clinical significance:

circXPO1 had diagnostic value for MM (AUC 0.815) and high circXPO1 expression was associated with poor survival; positively related to blood beta2-MG levels and ISS stage

Description:

circXPO1 is up-regulated in multiple myeloma bone marrow and MM cell lines and is associated with worse survival. Functionally, circXPO1 promotes proliferation and cell-cycle progression and inhibits apoptosis, at least partly by sponging miR-495-3p to upregulate DDIT4, and its silencing suppresses xenograft tumor growth.

Confidence score:

0.8718
