circRNA-disease information
Expression pattern:
UP
Associated gene: lysine-specific histone demethylase 1A (LSD1), enhancer of zeste homolog 2 (EZH2), Phosphatase and tensin homolog (PTEN), epithelial membrane protein 1 (EMP1), H3K27me3, H3K4me2
Associated microRNA: -
Biological function: Circ100284 promotes osteosarcoma cell proliferation, viability, migration, invasion, cell-cycle progression, and is associated with tumor size, WHO grade, lung metastasis, and poorer survival; circ100284 inhibits apoptosis.
Molecular mechanism: Circ100284 directly binds LSD1 and EZH2 and epigenetically silences PTEN and EMP1, negatively regulating PTEN and EMP1 expression to exert its biological functions in osteosarcoma.
Biological pathway or process:

proliferation (promotes); migration (promotes); invasion (promotes); metastasis (promotes); apoptosis (inhibits); cell cycle (promotes); other pathway/process (other)

Detected method:
Q
Validation methods:

RT-qPCR; Clinical Sample Validation; Transfection; CCK8; Transwell Assay; Cell Cycle Assay; Annexin V/PI Flow Cytometry; RIP (RNA Immunoprecipitation); ChIP / ChIP-seq; Western Blot; Cohort Study; Survival Analysis

Clinical significance:

High circ100284 expression was positively correlated with tumor size, pathological stage, and lung metastasis, and negatively correlated with patient survival time; the circ100284 high-expression group had a poorer survival rate than the low-expression group.

Description:

Circ100284 is up-regulated in human osteosarcoma tissues and cell lines, and high expression is associated with larger tumors, higher WHO grade, lung metastasis, and poorer survival. Functionally, circ100284 promotes OS cell viability, proliferation, migration, invasion, and cell-cycle progression while suppressing apoptosis. Mechanistically, it binds LSD1 and EZH2 and epigenetically represses PTEN and EMP1 expression.

Confidence score:

0.7624
