circRNA-disease information
Expression pattern:
DN
Associated gene: TIMP3, DHX9, AGO2
Associated microRNA: miR-17-3p, miR-181b-5p
Biological function: Inhibits HCC growth, migration and metastasis in vitro and in vivo; acts as a tumor suppressor; lower expression is associated with aggressive clinicopathological characteristics and poor prognosis.
Molecular mechanism: DHX9 upregulation inhibits cSMARCA5 biogenesis; cSMARCA5 functions as a sponge for miR-17-3p and miR-181b-5p to upregulate TIMP3 and inhibit HCC growth and metastasis.
Biological pathway or process:

proliferation (inhibits); migration (inhibits); metastasis (inhibits); ceRNA regulation (promotes); other pathway/process (other)

Detected method:
Q
S
Validation methods:

Back-Splice Junction PCR / divergent primers PCR; RNase R Treatment; Sanger Sequencing; circRNA-seq; Actinomycin D / DRB Stability Assay; RT-qPCR; Northern Blot; FISH / smFISH; IHC (Immunohistochemistry); Nuclear-Cytoplasmic Fractionation; Clinical Sample Validation; RIP (RNA Immunoprecipitation); circRIP; RNA Pull-Down; Luciferase Reporter Assay; Transfection; CCK8; Transwell Assay; Wound Healing Assay; In Vivo Animal Model; H&E Staining; Western Blot; Cohort Study; Survival Analysis; ROC Analysis; Bioinformatics Analysis; RNA-seq

Clinical significance:

Downregulated cSMARCA5 is associated with aggressive clinicopathological characteristics, poorer overall survival and recurrence-free survival, and can serve as an independent prognostic marker after hepatectomy.

Description:

cSMARCA5 (hsa_circ_0001445), derived from SMARCA5 exons 15 and 16, is significantly downregulated in HCC and its low expression predicts aggressive disease and poor survival after hepatectomy. Functionally, cSMARCA5 suppresses HCC cell growth, migration and metastasis in vitro and in xenograft models. Mechanistically, DHX9 upregulation contributes to cSMARCA5 downregulation, while cSMARCA5 sponges miR-17-3p and miR-181b-5p to upregulate the tumor suppressor TIMP3.
