circRNA-disease information
Expression pattern:
DN
Associated gene: HNRNPD
Associated microRNA: -
Biological function: not specified
Molecular mechanism: HNRNPD suppresses circCDK1 biogenesis and shifts the linear splicing/backsplicing balance toward CDK1 mRNA, thereby affecting CDK1 mRNA/protein levels linked to cell cycle and apoptosis phenotypes; circCDK1 itself showed no direct effect on cell cycle/apoptosis in tested assays.
Biological pathway or process:

cell cycle (other); apoptosis (other)

Detected method:
Q
S
Validation methods:

RT-qPCR; RNA-seq; circRNA-seq; Transfection; CCK8; Colony Formation Assay; Annexin V/PI Flow Cytometry; Cell Cycle Assay; Western Blot; IHC (Immunohistochemistry); In Vivo Animal Model; Clinical Sample Validation; Bioinformatics Analysis

Clinical significance:

circCDK1 is lower in ccRCC specimens than in para-ccRCC specimens, and the circCDK1:CDK1 mRNA ratio is lower in ccRCC tumour tissues than in para-ccRCC tissues.

Description:

In ccRCC, circCDK1 is decreased in tumor tissues, while HNRNPD modulates the linear splicing/backsplicing balance of the CDK1 locus. Altered circCDK1:CDK1 mRNA ratio reflects HNRNPD-dependent circRNA biogenesis control; circCDK1 itself showed no direct effect on cell cycle/apoptosis in functional assays reported.

Confidence score:

0.7373
