Biological function: circGSPT1 acts as a tumor suppressor in gastric carcinoma; it halts proliferation, migration, invasion, and autophagy in GC cells through its encoded protein GSPT1-238aa.
Molecular mechanism: circGSPT1 contains an IRES-driven spanning junction ORF that encodes GSPT1-238aa; GSPT1-238aa binds directly to vimentin and interacts with the vimentin/Beclin1/14-3-3 complex, modulating autophagy through inhibition of PI3K/AKT/mTOR signaling.
Biological pathway or process:

PI3K/AKT/mTOR (inhibits); autophagy (inhibits); proliferation (inhibits); migration (inhibits); invasion (inhibits); apoptosis (promotes)

Validation methods:

Back-Splice Junction PCR / divergent primers PCR; RNase R Treatment; Sanger Sequencing; RT-qPCR; RNA-seq; FISH / smFISH; Nuclear-Cytoplasmic Fractionation; Clinical Sample Validation; Co-IP; Luciferase Reporter Assay; Transfection; EdU Staining; Transwell Assay; Wound Healing Assay; Western Blot; IF (Immunofluorescence); Bioinformatics Analysis

Clinical significance:

circGSPT1 is significantly downregulated in GC tissues, and reduced expression is associated with lymphatic metastases; the encoded protein GSPT1-238aa is suggested as a novel target for GC treatment.

Description:

circGSPT1 is a human GSPT1-derived circular RNA that is downregulated in gastric carcinoma tissues and cell lines. It encodes the tumor suppressor peptide GSPT1-238aa through IRES-mediated translation, and this peptide binds the vimentin/Beclin1/14-3-3 complex to inhibit PI3K/AKT/mTOR signaling, autophagy, proliferation, migration, and invasion in GC cells. Reduced circGSPT1 expression is associated with lymphatic metastases, supporting its potential clinical relevance in GC.
