suppresses migration/invasion/metastasis and glycolysis in LUAD cells |
| Molecular mechanism: |
circDCUN1D4 binds HuR and promotes HuR cytoplasmic export; acts as a scaffold to facilitate HuR binding to TXNIP mRNA and stabilizes TXNIP mRNA; also directly base-pairs with TXNIP 3′UTR forming a circDCUN1D4/HuR/TXNIP ternary complex |

| Clinical significance: |
Downregulated circDCUN1D4 was more prevalent in lymph node metastatic tissues and was an independent risk factor for overall survival; low circDCUN1D4 expression had shorter OS. |
| Description: |
In LUAD, circDCUN1D4 is down-regulated and functions as a tumor suppressor by inhibiting metastasis-related behaviors and glycolysis. Mechanistically, it binds HuR, promotes HuR cytoplasmic export, and stabilizes TXNIP mRNA (also via direct interaction with TXNIP 3′UTR), forming a circDCUN1D4/HuR/TXNIP complex. Low circDCUN1D4 expression is associated with lymph node metastasis and worse overall survival. |
