circRNA-disease information
Expression pattern:
UP
Associated gene: LSD1, EZH2, KLF2, APC
Associated microRNA: -
Biological function: CircLRP6 promotes osteosarcoma cell proliferation, migration and invasion, inhibits apoptosis, promotes cell cycle progression, and acts as an oncogene in OS.
Molecular mechanism: CircLRP6 binds LSD1 and EZH2 and mediates their binding to promoter regions of KLF2 and APC, thereby inhibiting KLF2 and APC expression at the transcriptional level.
Biological pathway or process:

proliferation (promotes); migration (promotes); invasion (promotes); apoptosis (inhibits); cell cycle (promotes); Wnt/beta-catenin (promotes)

Detected method:
Q
Validation methods:

RT-qPCR; Clinical Sample Validation; Nuclear-Cytoplasmic Fractionation; RIP (RNA Immunoprecipitation); ChIP / ChIP-seq; Transfection; CCK8; Annexin V/PI Flow Cytometry; Cell Cycle Assay; Transwell Assay; Western Blot; Cohort Study; Survival Analysis; ROC Analysis

Clinical significance:

CircLRP6 has diagnostic potential for OS, and high circLRP6 expression is associated with shorter disease-free survival and overall survival; circLRP6 expression is an independent risk factor for OS prognosis.

Description:

CircLRP6 is up-regulated in osteosarcoma tissues and cell lines and high expression predicts poor survival. Functionally, circLRP6 promotes OS cell proliferation, migration, invasion and cell cycle progression while inhibiting apoptosis. Mechanistically, it binds LSD1 and EZH2 to suppress tumor-suppressor genes KLF2 and APC, contributing to OS development.
