circRNA-disease information
Expression pattern:
DN
Associated gene: PTEN, PI3K, AKT, ERK, p53, p21, E-cadherin, N-cadherin, vimentin, ZEB1, cleaved caspase-3, cleaved caspase-8, cleaved caspase-9
Associated microRNA: miR-21
Biological function: circHIAT1 functions as a tumor inhibitor in gastric cancer cells; its overexpression suppresses cell proliferation and EMT, induces apoptosis, decreases cell viability, and inhibits PTEN/PI3K/AKT and ERK pathway activation.
Molecular mechanism: circHIAT1 negatively regulates miR-21, thereby increasing PTEN and suppressing PI3K/AKT and ERK signaling; miR-21 mimic reverses the growth-inhibitory, pro-apoptotic and EMT-suppressive effects of circHIAT1 overexpression.
Biological pathway or process:

proliferation (inhibits); apoptosis (promotes); EMT (inhibits); PI3K/AKT (inhibits); ERK (inhibits); p53 signaling (promotes)

Detected method:
Q
Validation methods:

RT-qPCR; Clinical Sample Validation; Transfection; CCK8; Annexin V/PI Flow Cytometry; Western Blot

Clinical significance:

circHIAT1 could be a novel target for GC treatment and a promising effective therapeutic target in GC.

Description:

In this study, circHIAT1 was down-regulated in gastric cancer tissues and acted as a tumor suppressor in MKN28 and MKN45 gastric cancer cells. circHIAT1 overexpression inhibited proliferation and EMT, promoted apoptosis, and suppressed PTEN/PI3K/AKT and ERK signaling by downregulating miR-21, suggesting therapeutic potential in gastric cancer.
