| Expression pattern: |
UP |
| Associated gene: |
SLC7A5, PD-L1, AGO2 |
| Associated microRNA: |
miR-744-5p |
| Biological function: |
Promotes GC cell proliferation and immune escape; attenuates the anti-tumor effect of anti-PD-L1 in vivo; circSCUBE3 knockdown suppresses GC immune escape and can enhance the anti-tumor effect of PD-L1-neutralizing antibody. |
| Molecular mechanism: |
circSCUBE3 acts as a miR-744-5p sponge to upregulate SLC7A5, thereby regulating PD-L1 expression and affecting GC immune escape and anti-PD-L1 response. |

| Validation methods: |
RT-qPCR; Bioinformatics Analysis; Clinical Sample Validation; Actinomycin D / DRB Stability Assay; RNase R Treatment; Transfection; Colony Formation Assay; EdU Staining; Flow Cytometry(Non-apoptosis/cycle); ELISA; RIP (RNA Immunoprecipitation); Luciferase Reporter Assay; Western Blot; In Vivo Animal Model; Cohort Study; Survival Analysis |
| Clinical significance: |
circSCUBE3 high expression is associated with lymph node metastasis, TNM staging, and worse 5-year survival in GC patients; targeted knockdown may enhance the efficacy of PD-L1-neutralizing antibodies. |
| Description: |
circSCUBE3 is an up-regulated human circRNA in gastric cancer derived from the SCUBE3 gene. It promotes GC cell proliferation and immune escape through the circSCUBE3/miR-744-5p/SLC7A5 axis, regulates PD-L1 expression, and reduces the anti-tumor activity of anti-PD-L1 treatment in vivo. High circSCUBE3 expression is associated with lymph node metastasis, TNM staging, and worse 5-year survival. |
