Molecular mechanism: R-2HG induces circRNA-51217 generation, partly associated with increased m6A level of TGFbeta1 mRNA; circRNA-51217 acts as a miRNA sponge for miRNA-646, relieving repression of TGFbeta1 and activating TGFbeta1/p-Smad2/3 signaling. AR suppresses this axis by regulating TGFbeta1 transcription and ADAR2-mediated circRNA generation.

Description:

circRNA-51217 is induced by R-2HG/IDH1 R132H-related conditions in prostate cancer cells with negative or low AR expression. It promotes PCa cell invasion by sponging miRNA-646, increasing TGFbeta1 expression, and activating TGFbeta1/p-Smad2/3 signaling; AR counteracts this pathway partly through TGFbeta1 transcriptional repression and ADAR2-mediated suppression of circRNA generation.
