circRNA-disease information
Expression pattern:
UP
Associated gene: LY6E, AGO2
Associated microRNA: miR-637
Biological function: Promotes migration, invasion, proliferation, tumor growth and metastasis of cholangiocarcinoma cells; inhibits apoptosis; exosomal circ-0000284 stimulates migration and proliferation of surrounding normal cells and suppresses their apoptosis.
Molecular mechanism: circ-0000284 acts as a ceRNA by sponging miR-637 to up-regulate LY6E expression; circ-0000284 can also be transferred via exosomes from cholangiocarcinoma cells to surrounding normal cells.
Biological pathway or process:

ceRNA regulation (promotes); proliferation (promotes); migration (promotes); invasion (promotes); metastasis (promotes); apoptosis (inhibits)

Detected method:
Q
Validation methods:

RT-qPCR; RNase R Treatment; Sanger Sequencing; Clinical Sample Validation; Transfection; EdU Staining; Transwell Assay; Annexin V/PI Flow Cytometry; Luciferase Reporter Assay; Nuclear-Cytoplasmic Fractionation; RIP (RNA Immunoprecipitation); FISH / smFISH; Bioinformatics Analysis; Western Blot; In Vivo Animal Model; H&E Staining; IHC (Immunohistochemistry)

Clinical significance:

High expression of circ-0000284 in tumor tissues was associated with grade of differentiation, TNM stage and lymph node invasion; circ-0000284 was elevated in plasma exosomes of cholangiocarcinoma patients compared with healthy controls.

Description:

circ-0000284 is up-regulated in cholangiocarcinoma cell lines, tumor tissues and plasma exosomes. It promotes cholangiocarcinoma cell proliferation, migration, invasion, tumor growth and metastasis while inhibiting apoptosis through the miR-637/LY6E ceRNA axis. Exosome-transmitted circ-0000284 can also be delivered to surrounding normal cells and promote malignant-like cellular behaviors.

Confidence score:

0.8511
