circRNA-disease information
Expression pattern:
DN
Associated gene: APC, GATA3, LZTS1, MAPK10, SMAD5
Associated microRNA: miR-135b-5p
Biological function: circ_0000977 is down-regulated in TNBC and may act as a tumor-suppressive, discriminative molecular signature and cell-free diagnostic biomarker; lower expression is associated with shorter disease-free survival, younger age, and larger tumor size.
Molecular mechanism: Potential ceRNA mechanism in which circ_0000977 sponges miR-135b-5p and regulates downstream mRNAs, especially APC and GATA3, in TNBC.
Biological pathway or process:

ceRNA regulation (promotes); Wnt/beta-catenin (inhibits); other pathway/process (other)

Detected method:
Q
M
Validation methods:

Back-Splice Junction PCR / divergent primers PCR; Sanger Sequencing; RT-qPCR; Microarray; Clinical Sample Validation; Transfection; Cohort Study; Survival Analysis; ROC Analysis; Bioinformatics Analysis

Clinical significance:

Cell-free circulating circ_0000977 in plasma is a potential diagnostic biomarker for TNBC; low circ_0000977 expression is associated with shorter DFS, larger tumors, and younger patients.

Description:

This study identified circ_0000977/hsa_circ_0000977 as a down-regulated circRNA in TNBC compared with non-TNBC tumors and validated this pattern in tumor tissues, cell lines, and plasma. The authors propose that circ_0000977 participates in a ceRNA network by interacting with miR-135b-5p and regulating downstream tumor-suppressor mRNAs, especially APC and GATA3. Clinically, plasma circ_0000977 showed diagnostic potential for TNBC, and lower tumor expression was associated with shorter disease-free survival.
