circRNA-disease information
Expression pattern:
UP
Associated gene: NOVA2
Associated microRNA: hsa-miR-7-5p / miR-7
Biological function: circ-U2AF1 acts as an oncogene in glioma, promoting glioma cell proliferation, migration, invasion, metastasis-like phenotypes and tumorigenesis, while inhibiting apoptosis and affecting cell cycle progression.
Molecular mechanism: circ-U2AF1 functions as a competing endogenous RNA that sponges hsa-miR-7-5p, thereby derepressing and upregulating NOVA2 to promote glioma malignancy.
Biological pathway or process:

ceRNA regulation (promotes); proliferation (promotes); migration (promotes); invasion (promotes); metastasis (promotes); apoptosis (inhibits); cell cycle (other); p53 signaling (inhibits)

Detected method:
Q
Validation methods:

RT-qPCR; Clinical Sample Validation; Bioinformatics Analysis; Transfection; MTT; Annexin V/PI Flow Cytometry; Cell Cycle Assay; Transwell Assay; Luciferase Reporter Assay; RNA Pull-Down; Western Blot; In Vivo Animal Model

Clinical significance:

circ-U2AF1 expression was positively correlated with glioma grade and circ-U2AF1 was suggested as a prognostic biomarker and therapeutic target.

Description:

circ-U2AF1 (hsa_circ_0061868) is upregulated in human glioma tissues and glioma cell lines and its expression positively correlates with glioma grade. Functionally, circ-U2AF1 promotes glioma cell proliferation, migration, invasion, metastasis-like behavior and xenograft tumor growth, while suppressing apoptosis. Mechanistically, it sponges hsa-miR-7-5p to derepress NOVA2, forming a circ-U2AF1/hsa-miR-7-5p/NOVA2 regulatory pathway with potential prognostic and therapeutic relevance.

Confidence score:

0.705
