circRNA-disease information
Expression pattern:
UP
Associated gene: IGF2BP2, STAT3, IGF2BP1, GRWD1, NEU, SLC7A5, TRAPPC9, TP53
Associated microRNA: miR-1207-5p
Biological function: regulates gene expression and key factors for cell proliferation; affects proliferative/cell cycle-related pathways; functions as a ceRNA for IGF2BP2 and modulates STAT3 and p53 levels
Molecular mechanism: circHIPK3 contains an 11-mer motif that binds IGF2BP2; circHIPK3 competes with target mRNAs (e.g., STAT3) for IGF2BP2 binding and influences target mRNA stability; circHIPK3 can nucleate multiple IGF2BP2 molecules and promote IGF2BP2 condensate formation (potentially via phase separation).
Biological pathway or process:

proliferation (other); cell cycle (other); ceRNA regulation (other); p53 signaling (other)

Detected method:
Q
S
Validation methods:

RNA-seq; RT-qPCR; Back-Splice Junction PCR / divergent primers PCR; RIP (RNA Immunoprecipitation); Luciferase Reporter Assay; Transfection; Western Blot; Northern Blot; RNase R Treatment; IF (Immunofluorescence); Cell Cycle Assay; Survival Analysis; Cohort Study; Bioinformatics Analysis; Clinical Sample Validation

Clinical significance:

High circHIPK3 expression is positively associated with overall survival in BC patients; circHIPK3 is upregulated in NMIBC compared to MIBC.

Description:

In bladder cancer cells, circHIPK3 binds the RBP IGF2BP2 via an internal 11-mer motif and modulates IGF2BP2 availability to target mRNAs (e.g., STAT3), affecting target mRNA stability and proliferation/cell-cycle related programs. circHIPK3 can nucleate multiple IGF2BP2 molecules to form cytoplasmic condensates. Clinically, higher circHIPK3 expression is associated with better overall survival in NMIBC cohorts and differs between NMIBC and MIBC.

Confidence score:
