circRNA-disease information
Expression pattern:
UP
Associated gene: E2F2, c-Myc
Associated microRNA: miR-125 family, miR-125a, miR-125b, let-7b
Biological function: Promotes proliferation of gastric cancer cells; silencing circPVT1 suppresses growth, colony formation, and DNA synthesis.
Molecular mechanism: circPVT1 acts as a miRNA sponge for miR-125 family members, inhibits miR-125 activity, facilitates E2F2 expression, and may facilitate c-Myc expression by acting as a sponge for let-7b; its upregulation is partly due to amplification of its genomic locus.
Biological pathway or process:

proliferation (promotes); ceRNA regulation (promotes); other pathway/process (other)

Detected method:
Q
S
Validation methods:

Back-Splice Junction PCR / divergent primers PCR; RNase R Treatment; Sanger Sequencing; Actinomycin D / DRB Stability Assay; RT-qPCR; RNA-seq; Nuclear-Cytoplasmic Fractionation; Clinical Sample Validation; Luciferase Reporter Assay; Transfection; CCK8; EdU Staining; Colony Formation Assay; Cohort Study; Survival Analysis; ROC Analysis; Bioinformatics Analysis

Clinical significance:

circPVT1 is an independent prognostic marker for overall survival and disease-free survival in gastric cancer; patients with low circPVT1 levels had shorter OS and DFS, and combined circPVT1 expression with TNM stage improved prognostic value.

Description:

circPVT1 is a PVT1-derived human circRNA that is frequently upregulated in gastric cancer tissues, partly due to genomic DNA amplification at its locus. It promotes gastric cancer cell proliferation by functioning as a cytoplasmic miRNA sponge, especially for the miR-125 family, with downstream effects including E2F2 and possibly c-Myc regulation. Clinically, higher circPVT1 expression is associated with better OS and DFS and serves as an independent prognostic marker in gastric cancer.
