circRNA-disease information
Expression pattern:
UP
Associated gene: PDL1
Associated microRNA: miR-511-3p
Biological function: Promotes NSCLC cell viability, proliferation, invasion, EMT, angiogenesis, immune escape, and xenograft tumor growth; inhibits apoptosis.
Molecular mechanism: circ_0003528 functions as a cytoplasmic miR-511-3p sponge and increases PDL1 expression, thereby promoting malignant transformation and immune escape in NSCLC.
Biological pathway or process:

proliferation (promotes); apoptosis (inhibits); invasion (promotes); EMT (promotes); angiogenesis (promotes); immune regulation (promotes); ceRNA regulation (other)

Detected method:
Q
Validation methods:

RNase R Treatment; RT-qPCR; Nuclear-Cytoplasmic Fractionation; Clinical Sample Validation; RNA Pull-Down; Luciferase Reporter Assay; Transfection; MTT; EdU Staining; Annexin V/PI Flow Cytometry; Transwell Assay; Tube Formation Assay; ELISA; Western Blot; IHC (Immunohistochemistry); In Vivo Animal Model; Survival Analysis; Bioinformatics Analysis

Clinical significance:

High circ_0003528 expression was associated with worse prognosis and shorter 5-year overall survival in NSCLC patients; it may serve as a therapeutic target for NSCLC.

Description:

In this study, circ_0003528 was up-regulated in NSCLC tissues and cell lines and high expression predicted worse prognosis. Functionally, circ_0003528 promoted malignant transformation, angiogenesis, immune escape, and xenograft tumor growth while suppressing apoptosis. Mechanistically, it acted as a cytoplasmic sponge for miR-511-3p to increase PDL1 expression, forming a circ_0003528/miR-511-3p/PDL1 regulatory axis.

Confidence score:

0.8354
