| Expression pattern: |
DN |
| Associated gene: |
AGO2, RASGEF1B, Ras/MAPK signaling pathway |
| Associated microRNA: |
miR-3194-5p |
| Biological function: |
circDHPR suppresses HCC cell proliferation, colony formation, invasion, migration, EMT, tumorigenesis and lung metastasis; it induces G0/G1 phase cell cycle arrest. |
| Molecular mechanism: |
circDHPR acts as a ceRNA sponge for miR-3194-5p, thereby upregulating RASGEF1B and suppressing the Ras/MAPK signaling pathway. |
| Biological pathway or process: |
MAPK (inhibits); proliferation (inhibits); migration (inhibits); invasion (inhibits); metastasis (inhibits); EMT (inhibits); cell cycle (other); ceRNA regulation (promotes) |

| Validation methods: |
Back-Splice Junction PCR / divergent primers PCR; RNase R Treatment; Sanger Sequencing; Actinomycin D / DRB Stability Assay; RT-qPCR; Microarray; FISH / smFISH; Clinical Sample Validation; RIP (RNA Immunoprecipitation); Luciferase Reporter Assay; Transfection; CCK8; Colony Formation Assay; Cell Cycle Assay; Transwell Assay; Wound Healing Assay; In Vivo Animal Model; H&E Staining; IHC (Immunohistochemistry); Western Blot; Cohort Study; Survival Analysis; Bioinformatics Analysis |
| Clinical significance: |
Low circDHPR expression was associated with poor overall survival and disease-free survival, advanced T stage, and poorer clinicopathological features; circDHPR may act as an independent prognostic marker and biomarker/therapeutic target for HCC. |
| Description: |
circDHPR is downregulated in HCC and functions as a tumor suppressor. Its overexpression inhibits proliferation, invasion, migration, EMT, tumor growth and lung metastasis by sponging miR-3194-5p, upregulating RASGEF1B, and suppressing the Ras/MAPK pathway. Clinically, higher circDHPR expression is associated with better OS and DFS, suggesting prognostic and therapeutic potential. |
