circRNA-disease information
Expression pattern:
UP
Associated gene: KRAS
Associated microRNA: miR-101-3p
Biological function: promotes proliferation, cell cycle progression, and aerobic glycolysis; inhibits apoptosis; promotes tumor growth in vivo
Molecular mechanism: circ-MEMO1 acts as a miRNA sponge for miR-101-3p to up-regulate KRAS, promoting glycolysis-related enzymes (HK2, LDHA) and malignant phenotypes
Biological pathway or process:

proliferation (promotes); cell cycle (promotes); apoptosis (inhibits); glycolysis (promotes); ceRNA regulation (promotes)

Detected method:
Q
Validation methods:

RT-qPCR; RNase R Treatment; Actinomycin D / DRB Stability Assay; Nuclear-Cytoplasmic Fractionation; Transfection; MTT; Cell Cycle Assay; Annexin V/PI Flow Cytometry; Luciferase Reporter Assay; RIP (RNA Immunoprecipitation); RNA Pull-Down; Western Blot; In Vivo Animal Model; Clinical Sample Validation; Survival Analysis; ROC Analysis; Bioinformatics Analysis

Clinical significance:

High expression of circ-MEMO1 predicted poor prognosis in NSCLC patients; serum exosomal circ-MEMO1 (exo-circ-MEMO1) may be a diagnostic marker (ROC AUC ~0.76).

Description:

circ-MEMO1 is up-regulated in NSCLC tissues and serum exosomes and functions as an oncogenic circRNA. It mainly localizes to the cytoplasm and promotes NSCLC cell proliferation, cell-cycle progression, and aerobic glycolysis while inhibiting apoptosis, largely via the circ-MEMO1/miR-101-3p/KRAS ceRNA axis. High circ-MEMO1 correlates with poor survival and exosomal circ-MEMO1 shows diagnostic potential by ROC analysis.

Confidence score:

0.8709
