circRNA-disease information
Expression pattern:
DN
Associated gene: RACK1, OGT, Galectin-1
Associated microRNA: -
Biological function: inhibits lymphangiogenesis, invasion, tumor growth, and lymph node metastasis in cervical cancer
Molecular mechanism: circVPRBP binds RACK1 (WD3 region) and blocks S122 O-GlcNAcylation by reducing OGT-RACK1 interaction, promoting RACK1 ubiquitination/proteasomal degradation, thereby decreasing Galectin-1 and pAKT/pFAK signaling to inhibit lymphangiogenesis and LN metastasis
Biological pathway or process:

lymphangiogenesis (inhibits); invasion (inhibits); proliferation (inhibits); metastasis (inhibits); PI3K/AKT (inhibits); ubiquitination (promotes)

Detected method:
Q
H
Validation methods:

Back-Splice Junction PCR / divergent primers PCR; Sanger Sequencing; RT-qPCR; RNase R Treatment; Actinomycin D / DRB Stability Assay; Nuclear-Cytoplasmic Fractionation; ISH (In Situ Hybridization); FISH / smFISH; Clinical Sample Validation; Cohort Study; Survival Analysis; Transfection; Transwell Assay; Tube Formation Assay; CCK8; Colony Formation Assay; EdU Staining; Western Blot; RNA Pull-Down; RIP (RNA Immunoprecipitation); Co-IP; In Vivo Animal Model; IHC (Immunohistochemistry); H&E Staining

Clinical significance:

Loss/low expression of circVPRBP is associated with LN metastasis and shorter overall survival and disease-free survival; circVPRBP is an independent prognostic factor

Description:

circVPRBP (hsa_circ_0065898) is down-regulated in cervical cancer, especially in lymph node metastatic cases, and low expression correlates with worse survival. Functionally, circVPRBP suppresses invasion, lymphangiogenesis, tumor growth and lymph node metastasis by binding RACK1 and blocking its S122 O-GlcNAcylation, promoting ubiquitin-proteasome degradation of RACK1 and reducing Galectin-1/pAKT/pFAK signaling.

Confidence score:

0.8905
