| Expression pattern: |
DN |
| Associated gene: |
Ago2 |
| Associated microRNA: |
miR-382-5p |
| Biological function: |
Inhibits LPS-induced ferroptosis and inflammatory response; promotes cell viability in HPAEpiCs under sepsis-induced ALI conditions. |
| Molecular mechanism: |
Acts as a ceRNA/miRNA sponge for miR-382-5p; negatively regulates miR-382-5p, and miR-382-5p overexpression reverses circ-PRKCI protective effects. |
| Biological pathway or process: |
ferroptosis (inhibits); inflammation (inhibits); proliferation (promotes) |

| Validation methods: |
RT-qPCR; RNase R Treatment; Actinomycin D / DRB Stability Assay; Luciferase Reporter Assay; RIP (RNA Immunoprecipitation); Transfection; CCK8; ELISA; ROC Analysis; Bioinformatics Analysis; Clinical Sample Validation |
| Clinical significance: |
circ-PRKCI differentiates septic ALI patients from healthy individuals and septic non-ALI patients with high AUC (0.996 and 0.999), suggesting diagnostic biomarker potential. |
| Description: |
circ-PRKCI is down-regulated in septic ALI patient plasma and in LPS-treated HPAEpiCs. Its overexpression protects HPAEpiCs by suppressing ferroptosis and inflammatory cytokine release, mainly by sponging and negatively regulating miR-382-5p. circ-PRKCI shows strong diagnostic potential for septic ALI based on ROC analysis. |
