circRNA-disease information
Expression pattern:
UP
Associated gene: IGF2BP1, METTL3, G3BP1, m6A
Associated microRNA: -
Biological function: promotes hypoxia-induced neuronal apoptosis; contributes to penumbral injury; associated with synaptic damage
Molecular mechanism: m6A-dependent binding of IGF2BP1 to CircOGDH at splice junction motifs; stabilization by stress granule recruitment/compartmentalization; post-transcriptional regulation of CircOGDH stability
Biological pathway or process:

apoptosis (promotes); mitochondrial function (other); other pathway/process (other)

Detected method:
Q
Validation methods:

RT-qPCR; RNase R Treatment; RIP (RNA Immunoprecipitation); MeRIP / MeRIP-seq; RNA Pull-Down; FISH / smFISH; Luciferase Reporter Assay; Actinomycin D / DRB Stability Assay; Transfection; Co-IP; Western Blot; IF (Immunofluorescence); In Vivo Animal Model; TUNEL; Bioinformatics Analysis; Clinical Sample Validation

Clinical significance:

penumbra-specific transcript with diagnostic and therapeutic relevance; associated with infarct volume and penumbra size in rodent stroke models

Description:

CircOGDH is up-regulated in hypoxic neurons/ischemic penumbra and becomes enriched with m6A. IGF2BP1 binds m6A-modified CircOGDH and recruits it into stress granules to stabilize it, thereby promoting hypoxia-associated neuronal injury/apoptosis in acute ischemic stroke models.

Confidence score:

0.8355
