circRNA-disease information
Expression pattern:
UP
Associated gene: TET2, AGO2
Associated microRNA: miR-211-5p
Biological function: promotes lipolysis and white adipose browning (thermogenic remodeling) in adipocytes; promotes adipose wasting in vivo
Molecular mechanism: Acts as a miRNA sponge (ceRNA) for miR-211-5p to relieve repression of TET2; interaction supported by luciferase reporter, RIP (AGO2), and FISH co-localization
Biological pathway or process:

lipid metabolism (promotes); proliferation (not specified); ceRNA regulation (promotes); mitochondrial function (promotes)

Detected method:
Q
S
Validation methods:

RNA-seq; Bioinformatics Analysis; RT-qPCR; RNase R Treatment; divergent primers PCR; Sanger Sequencing; Nuclear-Cytoplasmic Fractionation; FISH / smFISH; RIP (RNA Immunoprecipitation); Luciferase Reporter Assay; Transfection; Western Blot; Oil Red O Staining; ELISA; IF (Immunofluorescence); In Vivo Animal Model; H&E Staining; ROC Analysis; Survival Analysis; Clinical Sample Validation; Cohort Study

Clinical significance:

circAMOTL1 expression positively correlated with weight loss severity and showed diagnostic performance for cachexia (ROC AUC=0.826).

Description:

circAMOTL1 (hsa_circ_0000350) is up-regulated in subcutaneous adipose tissue of cachectic cancer patients and correlates with weight loss severity. It promotes adipocyte lipolysis and WAT browning by sponging miR-211-5p, which relieves repression of TET2, and it exacerbates adipose wasting in a C26 tumor-bearing mouse cachexia model. It also shows diagnostic value (ROC) and high expression is associated with worse overall survival.

Confidence score:

0.8846
